About
This study aims to investigate the shared and disease-specific proteomic features of three common upper airway diseases: obstructive sleep apnea (OSA), allergic rhinitis (AR), and chronic rhinosinusitis (CRS). These conditions are prevalent, often comorbid, and associated with substantial morbidity, yet their molecular pathogenesis remains poorly defined. Our central research question is: what are the circulating protein signatures associated with each disease, and how do they overlap or differ across disease groups? To address this, we will use the UK Biobank Pharma Proteomics Project (UKB-PPP) dataset, which provides Olink-based serum protein measurements in a large, population-based cohort. We will define disease groups based on hospital diagnosis codes (ICD-10), self-reported symptoms, and medication data, and compare proteomic profiles between cases and matched controls. Statistical analyses will include multivariable regression, cluster analysis, and pathway enrichment to identify biologically meaningful patterns. We will also explore whether specific proteins correlate with disease severity indicators such as BMI, comorbid asthma, or medication use. The ultimate objective is to improve understanding of the inflammatory and immunologic mechanisms underlying these conditions and to identify potential biomarkers that may aid in diagnosis, risk stratification, or treatment selection.1 Publication
